Research focus of the working group:

 

Rapid reconstitution of the adaptive immune system, and in particular its T cell arm, is critical for successful hematopoietic stem cell transplantation after radiation therapy or chemotherapy. Thus, our primary research interest is in lineage decisions during lymphoid development, with particular focus on T lineage differentiation. A better understanding of these mechanisms may ultimately lead to improved protocols for T lineage reconstitution after hematopoietic stem cell transplantation. Our previous work addressed the interplay of various cell signaling pathways during the αβ vs. γδ T lineage decision in the thymus, as well as the developmental plasticity of bone marrow derived precursor cells towards different signaling cues. These studies ultimately lead to the identification of a novel T cell progenitor population. Analysis of cellular and molecular determinants of thymus homing revealed that there is not one designated extrathymic T cell precursor. Rather the thymus itself provides a microenvironment imposing T lineage fate on entering precursor cells. Analysis of chemokine receptor deficient mice indicated that the capacity to enter the thymus critically depends on the chemokine receptors CCR7 and CCR9. Current work addresses the spatio-temporal regulation of extrathymic and intrathymic T cell differentiation using novel mouse models and state-of-the-art imaging technology. Furthermore, we study human hematopoietic precursors in order to gain insight into the efficacy of T lineage reconstitution of different human T cell precursor populations.
