Technical Equipment

The sorter lab maintains four high speed flow cytometry cell sorters (FACSAria Fusion and FACSAria IIu, Becton-Dickinson; MoFlo XDP upgrade and MoFlo XDP, Beckmann-Coulter).

 

Individual sorting speeds of these machines are comparable and mainly are individually determined by either sorting conditions or recommendations. For example, sorting cells with high purity and good yield is possible with a 70µm nozzle with a rate of app. 30.000 events/sec (equals 108 cells/hour).


All sorters offer the option to sort up to four cell populations in parallel as well as sorting a specific amount of cells into cell culturing plates (e.g. cloning setups) or onto a glass slide.


Aseptic sorting for the continued cultivation of cells is possible, but generally we recommend the addition of antibiotics to the culture of cells after sorting.

applicable fluorochromes:

  • Filter combinations for all usual fluorochromes are available (PerCP detection possible!)
  • Green-yellow laser specially suited for red fluorescent proteins, TexasRed and PE tandem dyes.

 special features:

  • The integration of the sorter into a Class II type safety cabinet combined with an additional aeorosol management provides optimal biosafety protection. The sorter is ideally suited for sorting of infectious material and of genetically modified cells of BSL 2.
  • ceramic nozzles in 70 µm, 85 µm, 100 µm and 130 µm sizes are available

applicable fluorochromes:

  • filter combinations for many usual fluorochromes are available (standard: 5 color e.g. FITC, PE, PECy5, PECy7, APC, APCCy7, TxRed);
  • Filter combinations for the simulataneous detection of GFP and YFP are available.
  • applications requiring the excitation in UV range are possible, e.g. SP- cells stained with Hoechst dye;
  • Attention: PerCP dye cannot be detected!

special features:

  • ceramic nozzle for many cell sizes: 70µm, 100µm, 150µm (more upon request), thereby specially suited for sorting large cells
